2017年9月底，FDA批准Verzenio (abemaciclib)用于治疗HR阳性和HER-2阴性晚期乳腺癌或转移性乳腺癌治疗。 Verzenio (abemaciclib)既可单独使用治疗化疗/内分泌疗法后转移性乳腺癌，也可与氟维司群(Fulvestrant)联用治疗HR+/HER2-的晚期或转移性乳腺癌患者。

Abemaciclib是一种CD4/6激酶抑制剂，这类激酶通过连接D-细胞周期蛋白而启动，在雌激素受体阳性（HR+）的乳腺癌细胞系中，细胞周期蛋白D1与CD4/6结合能视网膜母细胞瘤蛋白（Rb）的磷酸化、细胞周期进程和细胞增殖。在体外，Abemaciclib连续暴露会抑制Rb磷酸化并阻止从G1进入细胞周期的S期，导致细胞衰老和凋亡。在乳腺癌异种移植模型中，Abemaciclib单药或联合抗雌激素每天不间断给药能使肿瘤缩小。

ABEMACICLIB is an inhibitor of CD4/6 kinases that are activated by binding to D-cyclins that bind retinoblastoma protein (Rb) phosphorylation, cell cycle progression, and cell proliferation in estrogen-receptor-positive (HR+) breast cancer cell lines. In vitro, continuous exposure to Abemaciclib inhibits Rb phosphorylation and prevents entry from G1 into the S phase of the cell cycle, leading to cell senescence and apoptosis. In xenograft models of breast cancer, continuous daily administration of Abemaciclib alone or in combination with anti-estrogen resulted in tumor shrinkage.
